Optimising rAAV Purification: Thermo Scientific POROS™ CaptureSelect™ AAVX Resin Delivers High Capacity and Efficiency for Gene Therapy Manufacturing
Recombinant adeno-associated virus (rAAV) is a key tool for gene therapy, with applications ranging from rare diseases to widespread conditions like hemophilia. Production methods have standardized, with HEK293 cell transfection yielding high titers necessary for large-scale treatments. Thermo Scientific POROS™ CaptureSelect™ AAVX resin has become a commonly used approach for rAAV purification.
This study evaluates dynamic binding capacity (DBC) across different AAV serotypes and process conditions to optimize productivity, balancing efficiency for both clinical and commercial manufacturing scales.

- High binding capacity: AAVX resin achieves >1E15 capsids/mL at residence times ≥ 0.5 min for AAV2.
